The CRISPER project aims to develop a battery-operated, portable and modular diagnostic platform that integrates CRISPR/Cas technology with electrochemical biosensing. Within a One Health framework, the proposal targets the early detection of nucleic acid biomarkers across human, animal and environmental or food-related matrices, providing a versatile system to quantify DNA and RNA directly at the point of need through three different CRISPR/Cas toolkits based on the specific activation of Cas9, Cas12a and Cas13a. The modular platform combines sample preparation, nucleic acid amplification (using isothermal methods or PCR in a battery-powered thermocycler) and CRISPR-assisted electrochemical readout, converting them into clear ON/OFF electrical signals that are easy to interpret in low-infrastructure settings..
